LiveBox2 is a transparent chamber, developed for in-vitro models of physiological barriers (e.g. lung and intestinal
epithelium).

LiveBox2 is designed to simulate physiological barriers under both liquid-liquid (LLI) and air-liquid (ALI) interface conditions. It is equipped with two inputs and two outputs to ensure a flow of culture medium in both apical and basal environments. The 2 zones are divided by a porous membrane, suitably mounted on a removable holder.
The Clamp system supplied with LiveBox2 ensures the watertight closure of the system in both static and dynamic conditions (up to 0.5 mL/min in apical or basal compartments).

Features:

The system is designed to reproduce the typical seeding surface of a transwell insert for a 6-12-24 well plate.

Removable transparent glass bottom allows:
- real-time imaging during static/dynamic culture using inverted optical microscopy.

Removable Holder allows:
- the housing of several commercial porous membranes, depending on the specific cell type or tissue. Their material and porosity can be chosen to best simulate the desired reference transwell.
- easy and safe handling of the membrane on which the cells are sown
- post processing of the crop, including coloring or any other type of application.

Modularity: LiveBox2 can be connected with other cell culture systems to mimic cross talk between tissues (multi-organ simulation).

Configurations:

LiveBox2 -> 2 different configurations:
- tangential flow configuration, suitable for the simulation of physiological barriers.
The apical and basal chambers are perfused by 2 different and independent flows. The flow lines are parallel to the membrane surface.
- Sigmoidal flow configuration, suitable for perfusion of 3D cellular constructs or maintenance in floating conditions of cellular aggregates or spheroids. The flow lines pass through the chamber in vertical direction. Two sub-configurations can be implemented, based on the flow direction: top-down configuration (e.g. net perfusion of porous and thick constructs placed on the membrane) or bottom-up configuration (e.g. maintenance of spheroid under floating conditions in the basal chamber of LiveBox2).
