AUTOFOCUSED COUNTING IN 15 SECONDS

The automated cell counter LUNA II has integrated a novel focussing mechanism based on liquid lenses technology.

The liquid lenses do not use any mechanical moving parts to change the Z stage.

Instead, the position of the sample is rapidly obtained by the application of a small voltage to the liquid lens. The elimination of moving parts removes noise and significantly reduces the need of servicing. Even after multi-million cycles of focussing, the liquid lens does not require repair or service.

In combination with this novel focussing mechanism, the LUNA II has integrated a new autofocusing algorithm optimized for cell counting

The Luna-FL automated cell counter from Logos Biosystems is a quantum leap for automated cell counting and cell viability analysis.

The Luna-FLgives you sensitive and accurate live/dead cell counting results without limitations of cell types.

Unlike immortalized cell lines primary cells such as PBMCs splenocytes, neutrophils, and stem cells have been difficult to count with conventional cell counters such as coulter counter or image-based automated cell counters that utilize bright field optics.

Primary cells are often contaminated with undesirable debris, which can be confused with cells with conventional cell counters.

The Luna-FL is integrated with dual fluorescence microscope optics to overcome this problem.

Device has been easy to use, and simple for a new user. Autofocus works nicely which is important to us; we need consistent results because different people run cell counting as a first step of single cell analysis. Algorithm seems to work accurately as it differentiates between actual cells and cell debris or other particles. Counting with the 8-chamber slide is quite consistent, you just have to pay attention to even pipetting. Measuring with the 8-chamber slide is super easy and quick compared to Luna-stem that we used to use. Getting 8 counts and also saving all the files with two keystrokes frees you to do something else while the counter is running.

Challenge

Osteoporosis and degradation of bone mechanics affects millions of people each year, often leading to fractures and immobility. Therefore, regenerative medicine strategies are urgently needed. Bioprinting complex geometries with collagen, however, can be challenging and most scaffolds based on type I collagen and hydroxyapatite are fabricated by freeze-drying or casting, application that give bioengineers limited control over the final shape and porosity of their constructs.

Solution

The team at the Polytechnic University of Turin mitigated these challenges by combining the BIO X, a temperature controlled printhead and gelatin-supporting baths (the FRESH method) to 3D bioprint taller, more complex bone scaffolds with type I collagen.

Results

The study validates the potential for collagen nanocomposites as a material for high-resolution 3D bioprinting. The authors also suggest that using the hybrid material represents a significant step toward fabricating biomimetic patient-specific bone-like scaffolds on demand.

Challenge

Typically, gelatin, collagen, alginate and fibrin hydrogels are used in 3D bioprinting and tissue engineering because of their extracellular matrix (ECM) properties. However, the search for other natural ECM-like polymers has lagged behind. Chitosan, a natural, biocompatible and biodegradable polymer, has been approved by the FDA and EMA for use in wound dressings, but it is not stable under physiological conditions.

Solution

Conti et al. discovered that because of their ionic interactions and rheological properties, 3D bioprinting of chitosan and gamma-PGA structures together at certain concentrations of the biopolymer and crosslinker resulted in more favorable constructs. The CELLINK INKREDIBLE+ bioprinter was used to explore the bioprinting properties of this bioink formulation in order to compare it to other natural ECM material formulations.

Results

The 3D structures generated by the INKREDIBLE+ confirmed that the chitosan-gamma-PGA formulation is biocompatible and stable under shear stresses greater than that of human arteries and veins.

I'm testing the LUNA-II and it works great. I work for an ethanol plant and our operators count yeast. This will be a great way to streamline the yeast count results. It will also please our operators.

The best automatic digital imaging system
We need live cell time-lapse imaging for our studies but other imaging systems did not produce acceptable images for long-term imaging. The CELENA X is the only system that has given us satisfactory multi-fluorescence images of multi-well plates. The automatic laser focusing is especially fantastic. We don’t waste time taking individual images as the CELENA X does high-throughput imaging and focuses great for all wells over long periods of time. Image quality is great for every photo and making high-quality videos is easy. Cells remain healthy, and there is minimal fluorescence quenching even after extensive imaging (5 minute intervals for over 72 hours). I strongly recommend the CELENA X for live cell time-lapse imaging and other applications.

Use with larger samples (few liters)

The AmMag™ SA contains adapters for entering samples in 15 mL and 50 mL tubes. However, larger samples can be easily purified using the AmMag™ SA by simply collecting the magnetic beads from these samples after the binding step, and transferring the beads into 50 ml or 15 ml tubes. To help collect magnetic beads from larger samples, we provide the AmMag™ Wand and the AmMag™ Block.

Closed environment to minimize endotoxin contamination

The AmMag™ SA is equipped with air filters to clean incoming air, and further maintains a positive air flow through the instrument. These features prevent airborne endotoxins from entering the system and contaminating samples. To clean the instrument between runs, all tubing can be flushed and incubated for up to an hour with 0.1 N NaOH.

Complete washing and elution process automatically

After cultures have expressed the desired proteins or antibodies, magnetic beads are added to bind the targets. After this step, the cultures are transferred to 50 mL tubes and then moved to AmMag™ SA. The automated washing and elution steps purify the target proteins and antibodies from the magnetic beads.

The AmMag™ SA utilizes magnetic beads for purification. Since magnetic beads are free-flowing, cells and debris from cultures do not prevent them from binding target protein and antibodies.
